چکیده

This work examines protein adsorption on surfaces in an attempt to provide a fundamental understanding of the molecular-level mechanism for protein interactions with biomaterials. Carboxylic acid, amine, and hydroxyl terminated self-assembled monolayers (SAMs) are used to model negatively, positively, and hydrophilic/neutrally charged surfaces, respectively. The adsorption of fibrinogen and lysozyme on these SAM surfaces under different ionic strengths and pH values is studied using a surface plasmon resonance (SPR) sensor. It is shown that variations in ionic strength have little effect on the adsorbed amount of fibrinogen and lysozyme on these SAMs at a fixed pH value. Analysis of the effect of pH on protein adsorption at a fixed ionic strength shows that charge interactions are a significant component of protein adsorption. These studies provide insights into the role of hydrogen bonding and electrostatic interactions in protein adsorption.

Behenic Acid Monolayer and Bilayer Assemblies- A Study of Concanavalin A (Con A) Adsorption and its Interaction with Dextran Using Surface Plasmon Resonance Spectroscopy and Microscopy

Deposition of behenic acid (B.A) mono and bilayers onto gold coated surfaces was performed by Langmuir-Blodget dip-casting technique. Surface Plasmon resonance (SPR) and ellipsometry methods were employed for investigation of the monolayer and bilayer films. The adsorption of the biologically important molecule Concanavalin A (Con A) from bulk solution to these monolayers and bilayers as we...

Using Surface Plasmon Resonance Spectroscopy To Measure the Association of Detergents with Self-Assembled Monolayers of Hexadecanethiolate on Gold

This paper describes the use of surface plasmon resonance (SPR) spectroscopy to measure the rates and extents of association of four detergentsssodium dodecyl sulfate (SDS), â-octyl glucoside, Triton X-100, and Tween 20sto self-assembled monolayers (SAMs) of alkanethiolates on gold. SAMs presenting hexaethylene glycol groups resisted theadsorption of all four detergents. These samedetergents as...

Methods of reducing non-specific adsorption in microfluidic biosensors

Non-specific adsorption (NSA) of biomolecules is a persistent challenge in microfluidic biosensors. Microfluidic biosensors often have immobilized bioreceptors such as antibodies, enzymes, DNAs, etc, via linker molecules such as SAMs (self-assembled monolayers) to enhance immobilization. However, the linker molecules are very susceptible to NSA, causing false responses and decreasing sensitivit...

Fabrication and Characterization of the Fiber Optical Taper for a Surface Plasmon Resonance Sensor

For a fiber optical surface plasmon resonance (SPR) sensor a short part of its cladding should be removed to coat a thin layer of a metal. Usually this is problematic when an optical fiber with small core diameter is used. In this paper, a new method using µliter droplet of the HF acid for short fiber optical taper fabrication is reported. Using this method in a multi-mode optical fiber w...

Surface Plasmon Resonance Imaging Measurements of Electrostatic Biopolymer Adsorption onto Chemically Modified Gold Surfaces.

A combination of in situ and ex situ surface plasmon resonance (SPR) imaging experiments is used to characterize the differential electrostatic adsorption of proteins and synthetic polypeptides onto photopatterned monolayers at gold surfaces. The nonspecific electrostatic adsorption of proteins onto negatively charged self-assembled monolayers (SAMs) of 11-mercaptoundecanoic acid (MUA) is found...
